  • Calcitonin is a 32 amino acid polypeptide hormone that is produced in humans primarily by the Parafollicular (also known as C) cells of the thyroid, and in many other animals in the ultimobranchial body. It acts to reduce blood calcium Ca2+, opposing the effects of parathyroid hormone (PTH). It has been found in fish, reptiles, birds and mammals. Its importance in humans has not been as well established as its importance in other animals.
  • Calcitonin is formed by the proteolytic cleavage of a larger prepropeptide which is the product of the CALC1 gene (CALCA). The CALC1 gene belongs to a superfamily of related protein hormone precursors including islet amyloid precursor protein, calcitonin gene-related peptide, and the precursor of adrenomedullin. The hormone participates in calcium (Ca2+) and phosphorus metabolism. In many ways, calcitonin has the counter effects of parathyroid hormone (PTH).
  • Oral calcitonin may have a chondroprotective role in osteoarthritis (OA), according to data in rats presented last month at the 10th World Congress of the Osteoarthritis Research Society International (OARSI) in Boston, Massachusetts. Unlike several other potential disease-modifying OA drugs, calcitonin also has a “unique analgesic effect on bone pain, which might relieve at least in part symptoms accompanying joint disease,” says Dr. Karsdal, adding that this may be an important factor influencing future drug compliance.
  • This medication is used to treat certain bone problems (e.g., Paget's disease, postmenopausal osteoporosis) and to reduce high blood levels of calcium. Calcitonin is a man-made hormone that works by slowing bone loss and maintaining normal calcium levels in the blood. It may also help to reduce bone pain in people who have Paget's disease.
  • A joint venture between US-based Unigene Laboratories and China's Shijiazhuang Pharmaceutical Group will begin selling injectable calcitonin in China for the treatment of osteoporosis as of 1 April. China is the single largest osteoporosis market with an estimated 90 million sufferers and more than 15 million fractures per year, the company claimed. According to the Shanghai Medical Journal and Chinese medical experts, 90 per cent of Chinese citizens over 60, and virtually all of those over 70, have the disease. The Chinese government has identified osteoporosis as one of the top three health priorities in China. 
  • Unigene's net loss for the three months ended March 31, 2007 was $244,000, or $.00 per share, compared to a net loss of $3,243,000, or $.04 per share for the three months ended March 31, 2006.The three months ended March 31, 2007 and 2006 includes $393,000 and $165,000, respectively, in expenses for non-cash stock optio compensation. As of March 31, 2007, Unigene had approximately $697,000 in expenses for non-cash stock option compensation that remains to be recognized, primarily over the next 12 months. 
  • US pharmaceutical company Upsher-Smith Laboratories has been forced into staffing cuts and a restructuring exercise by delays in the US Food and Drug Administration's review of its nasal calcitonin product for osteoporosis. Upsher-Smith licensed the product, called Fortical, from nasal drug delivery specialist Unigene in 2002. This $10 million (€8.3m) agreement gave Upsher-Smith exclusive US rights to distribute and sell the finished product, with Unigene retaining manufacturing rights. 
  • An oral formulation of salmon calcitonin based on Emisphere Technologies' eligen delivery system has entered Phase III clinical trials for the treatment of osteoarthritis. According to Emisphere, the system has been used for oral delivery of a range of molecules varying in size from 500 Daltons to more than 150,000 Daltons. Oral formulations created with the eligen technology include tablets, solutions and capsules; Emisphere believes it is also applicable to controlled-release dosage forms. 
  • Miacalcic is registered in 97 countries around the world and it is estimated that it has been administered for over 3 million patient years since it was first launched in 1974 in an injectable form. Miacalcic Nasal Spray received its first approval in 1987. Its first European Union marketing licence for post-menopausal osteoporosis was obtained in Portugal in 1987, followed by subsequent approvals in other countries.
  • The osteoporosis pharmaceutical market is currently $7 billion and is expected to grow to $14 billion by 2011. According to IMS, a market research firm, worldwide sales of an existing Calcitonin-Salmon Nasal Spray were approximately $389 million in 2003, of which the U.S. accounts for an estimated $240 million.
