  • Furosemide (INN) or frusemide (former BAN) is a loop diuretic.
It is most commonly marketed by Sanofi-Aventis under the brand name Lasix.
  • The name of lasix is derived from lasts six (hours) -- referring to its duration of action.
  • Furosemide is a noncompetitive subtype-specific blocker of GABA-A receptors.
  • Furosemide is odorless white to slightly yellow crystalline powder.
  • Furosemide is a diuretic which is an anthranilic acid derivative.
It is practically insoluble in water, sparingly soluble in alcohol, freely soluble in dilute alkali solutions and insoluble in dilute acids.
  • Furosemide may undergo hydrolysis at sufficiently low pH.
The spherical agglomeration process is used to delay the dissolution of furosemide.
  • Hyperprostaglandin E syndrome/antenatal Bartter syndrome (HPS/aBS), a salt-losing tubulopathy of the furosemide type , is clinically characterized by polyhydramnios leading to premature delivery, hypoisosthenuria with marked polyuria, and severe hypercalciuria resulting
    in nephrocalcinosis.
  • Pre-systemic metabolism of frusemide is carried out by the intestine while systemic clearance of frusemide is mainly performed by the kidneys with contributions from the intestine and the liver.
  • The improving of the solubility and UV stability of furosemide, a poorly water soluble diuretic can be done by complexation with ß-cyclodextrin.
  • A sensitive, selective and efficient micellar liquid chromatographic (MLC) procedure was developed for the determination of furosemide (4-chloro-N-furfuryl-5-sulfamoylanthranilic acid) in urine samples by direct injection and UV detection.
  • Furosemide improves medullary oxygenation by reducing oxygen consumption rather than by increasing regional blood flow.
  • Frusemide is commonly used in conditions such as heart failure, where the pumping mechanism of the heart is less effective.
  • Frusemide is frequently used to facilitate fluid and electrolyte management of acute renal failure in many institutions.
Furosemide increases water content in renal tissue.
  • Urinary-system elimination of 18F-FDG can be mistaken for pathologic uptake. Furosemide helps eliminate this artifact.
  • Chronic subcutane-ous infusion of furosemide, with concomitant salt and water substitution, was used to examine the isolated effect of prolonged inhibition of NaCl transport in the cTALH/macula densa cells on expression of COX-2 in rat kidney cortex.
  • Frusemide has significant bronchodilatory effect when given through inhaled route but less effective than salbutamol.
  • NIR spectroscopy technique was used to evaluate 48 different formulations of furosemide tablets.
  • The short-term effects of intravenous administration of furosemide on key functions in the kidney cortex has been evaluated using magnetic resonance imaging (MRI) technique.
