- Chromatography is the ability to
separate molecules using partitioning characteristics of molecule to
remain in a stationary phase versus a mobile phase. Once a molecule is
separated from the mixture, it can be
isolated and quantified -
A mixture of various components enters a
chromatography process, and the different components are flushed through
the system at different rates. These differential rates of migration as
the mixture moves over adsorptive materials provide separation. Repeated
sorption/desorption acts that take place during the movement of the
sample over the stationary bed determine the rates. The smaller the
affinity a molecule has for the stationary phase, the shorter the time
spent in a column.

In paper chromatography, the mobile phase
is a solvent, and the stationary phase is water held in the fibres of
chromatography paper

In column chromatography, the mobile
phase is again a solvent, and the stationary phase is a finely divided
solid, such as silica gel or alumina

In thin layer chromatography, the mobile
phase is also a solvent, and the stationary phase is a thin layer of
finely divided solid,
such as silica gel or alumina, supported on glass or aluminium -
A gas is the mobile phase and the
stationary phase can be either a solid or a non- volatile liquid

Ion-exchange chromatography: separation
of proteins over a column filled with charged polymer beads . Positively
charged proteins bind to beads of negative charge & vice versa. Bound
proteins are eluted with salt. Least charged proteins will elute first
